Obez ve obez olmayan bireylerde total antioksidan kapasite (TAK), total oksidan seviye (TOS) ve tiyol disülfit ölçümü

Abstract

Bu çalışmadaki amacımız tiyol-disülfit dengesinin obeziteyle oluşan oksidatif strese etkisini araştırmaktır. Bu amaçla; yaşları 18 ile 65 arasında değişen, 45 sağlıklı bireyden (kontrol) ve 45 obez bireyden (deney) 2 grup oluşturuldu. Medipol Üniversitesi Mega Hastanesi Labaratuvarına başvuran bireylerden rutin tektikler için alınan serumlarda total antioksidan kapasite (TAK), total oksidan seviye (TOS) ve tiyol-disülfit dengesi, Erel ve Neşelioğlu tarafından geliştirilen metodla çalışıldı. Total kolesterol, trgigliserit, Low-Density lipoprotein kolesterol (LDL) ve High-Density Lipoprotein kolesterol (HDL) Cobas Roche 6000 otoanalizöründe enzimatik kolorimetrik yöntem ile çalışıldı. Obezitenin tanısında kullanılan BKİ hesaplaması bütün katılımcılara uygulandı. Buna göre 18,530 olan bireyler obez olarak tanımlandı. Kontrol grubunun BKİ ortalaması 21,8±1,7 iken deney grubunun BKİ ortalaması 34,5±3,9 olarak bulundu (p<0.01). Kontrol grubuyla karşılaştırıldığında deney grubun TG, TK ve LDL düzeyleri istatiksel olarak yüksek; HDL düzeyi ise istatiksel olarak düşük çıkmıştır (sırasıyla p<0.01, p<0.05, p<0.05, p<0.01). Kontrol grubuyla kaşılaştırıldığında deney grubunun TAK konsantrasyonu istatiksel olarak düşük; TOS konsantrasyonu ve OSİ değeri istatiksel olarak yüksek bulunmuştur (p<0.01). Kontrol grubuyla karşılaştırıldığında deney grubunun total tiyol ve serbest tiyol konsantrasyonu rakamsal daha düşük bulunsa da istatiksel açıdan bir fark bulunmamıştır (p>0,05). Kontrol grubuyla karşılaştırıldığında deney grubunun disülfit, disülfit/serbest tiyol, disülfit/total tiyol konsantrasyonu istatiksel olarak yüksek bulunmuştur (p<0.05). BKİ ile yaş, TK, TG, LDL, disülfit, disülfit/serbest tiyol ve disülfit/total tiyol pozitif korelasyon; HDL ve TAK negatif korelasyon göstermiştir (p<0.01). Sonuç olarak; obez bireylerde oksidatif stresin göstergesi olan parametreler yüksek bulunmuştur.The aim in this study is to investigate the effect of thiol-disulfide balance on the oxidative stress caused by obesity. For this purpose; 45 healthy individuals (control) and 45 obese individuals (experimental), aged between 18-65 years, were divided into two groups. The serums were taken for routine tests from individuals who applied to Medipol University Mega Hospital Laboratory. Total antioxidant capacity (TAC), total oxidant status (TOS) and thiol-disulfide balance in the serum were studied by the method developed by Erel and Neşelioğlu. Total cholesterol (TC), triglyceride (TG), Low-Density lipoprotein (LDL) and High-Density Lipoprotein (HDL) were studied by Cobas Roche 6000 autoanalyzer by enzymatic calorimetric method. Body mass index (BMI) calculation used in the diagnosis of obesity was applied to all participants. According to this, individuals with 18.530 were defined as obese. The mean BMI of the control group was 21.8±1.7; the mean BMI of the experimental group was 34.5±3.9 (p<0.01). The TC, TG and LDL levels of the experimental group were higher than control group; HDL levels of the experimental group were lower than control group (p<0.01, p<0.05, p<0.05, p<0.01 respectively). The TAC concentration of the experimental group was statistically significantly lower than control group; the TOS concentration and OSI value of the experimental group were statistically significantly higher than control group (p<0.01). The total thiol and native thiol concentrations of the experimental group were found to be lower than control group but there was no statistical difference (p>0,05). The concentration of disulfide, disulfide/native thiol, disulfide/total thiol in the experimental group were statistically significantly higher than control group (p<0.05). BMI correlated positively with age, TC, TG, LDL, disulfide, disulfide/native thiol and disulfide/total thiol; HDL and TAC showed negative correlation (p<0.01). As a result; the parameters that are indicative of oxidative stress in obese individuals were found to be high.
