Akım sitometri tekniği ile yeni doğan fare beyninden bölgesel olarak izole edilmiş spesifik hücrelerin protein profillerinin incelenmesi

Abstract

Memeli santral sinir sistemi (SSS) yapısal ve işlevsel organizasyonu ile yaşayan organizmalar arasında en karmaşık yapıya sahip olan sistemdir. Bu kompleks sistemin işleyiş mekanizması ise ancak beyni oluşturan hücrelerin, uzaysal ve zamansal konumları ile birlikte ele alınmasıyla anlaşılabilir. Çalışmamızda bu hücrelerin protein profillerinin çıkarılarak tanımlanması amaçlanmıştır. Bu amaçla; öncelikle yenidoğan farelerdeki hipokampüs ve korteks bölgelerine ait hücreler antikorlar ile işaretlenerek akım sitometri cihazı ile analiz edilmiştir. Analiz sonucu tespit edilen astrositler ile negatif seçilim yöntemi ile analize dâhil edilen nöronlar Fluorescence Activated Cell Sorting (FACS) tekniği ile ayrıştırılarak saflaştırılmıştır. Hipokampüs ve korteks bölgelerinden izole ettiğimiz nöron ve astrositlerdeki proteinlerin tanımlanması ve ekspresyonlarının belirlenmesi için kütle spektrometresi tabanlı protemik teknolojisi kullanılmıştır. Yenidoğan fare hipokampüsünden toplam 67, korteksten ise 107 protein tanımlanmıştır. Bu proteinlerden 45 tanesi hipokampüste, 85 tanesi kortekste bulunurken 22 tanesi her iki bölgede de tespit edilmiştir. Bunlara ek olarak, biyoinformatik yazılımlar kullanılarak tanımlanan proteinlerin sınıfları ve fonksiyonları incelenmiştir. Sonuç olarak bu çalışmada tanımlanan 22 ortak protein arasından 5 proteinin hipokampüs bölgesinde astrositlerde, korteks bölgesinde ise nöronlarda daha çok eksprese edildiği gözlenmiştir. Bu 5 proteinden 3'ünün özellikle migrasyon, bölünme ve maturasyon sürecinde rol oynadıkları belirlenmiştir.Central nervous system (CNS) of mammalians constitutes the most complex architecture among other living organisms in terms of structural and functional organization. The operation mechanism of this complex system could solely be deciphered by addressing the spatial and time-wise state of cells that compose brain. In our study, we aimed comprising protein profiles of these cells, along with obtaining their fulfilled identification. For this purpose, hippocampus and cortex cells of newborn mice were tagged with antibodies and analyzed by flow cytometry. Astrocytes that were detected by the analysis and neurons that were included in the analysis by negative selection method were both separated and purified by Fluorescence Activated Cell Sorting (FACS). In order to identify the proteins of neurons and astrocytes isolated from hippocampus and cortex regions and define their expression levels, mass spectrometry based proteomic technology was used. Total of 67 proteins from hippocampus and 107 proteins from cortex of newborn mice were identified. Among these proteins, 45 of them were detected in hippocampus, 85 of them were found in cortex and 22 of them were observed in both regions. In addition, identified proteins were screened in terms of their classes and functions by bioinformatics software. In conclusion, among 22 mutual proteins identified in this study, 5 were expressed densely in astrocytes in hippocampus and neurons in cortex. 3 of these 5 proteins were found to play crucial role in migration, division and maturation process.
