Obez sıçan modelinde ROUX-EN-Y gastrik bypass, whey proteini ve omega-3 yağ asidinin biyokimyasal parametrelere ve organ yağlanmasına etkisi

Abstract

Bu çalışmanın amacı; yüksek fruktozlu mısır şurubu verilerek obez yapılan sıçanlara uygulanan whey proteini ve omega-3 yağ asidi takviyesi ile Roux-en-Y gastrik bypass işleminin vücut ağırlığı kaybı, iç organlarda görülen histopatolojik değişimler ve biyokimyasal değişiklikler üzerindeki etkisini incelemektir. Çalışma kapsamında 28 adet Wistar albino dişi sıçan kullanılmış ve rastgele dört gruba ayrılmıştır. Tüm sıçanların sekiz hafta boyunca içme sularına %20 oranında yüksek fruktozlu mısır şurubu-55 eklenerek obez olmaları sağlanmıştır. Tüm sıçanlarda %40'tan fazla ağırlık artışı olduğu için morbid obez olarak değerlendirilmiştir. Dört hafta süresince oral gavaj yolu ile birinci gruba (kontrol) içme suyu, ikinci gruba whey proteini (0,5 g/kg/gün), üçüncü gruba omega-3 yağ asidi (300 mg/kg/gün) desteği ve dördüncü gruba Roux-en-Y gastrik bypass sonrası içme suyu verilmiştir. Çalışmanın başında, obezite gelişimi sonrasında ve çalışmanın sonunda her sıçandan 1 mL kan alınarak glikoz, toplam kolesterol, HDL-K, VLDL-K, AST, ALT ve ürik asit analizleri yapılmıştır. Çalışmanın sonunda, tüm sıçanların karaciğer, böbrek ve pankreası alınarak histopatolojik olarak değerlendirilmiştir. Vücut ağırlığında gruplar arası önemli fark görülmemiştir. Kontrol grubu hariç, diğer gruplarda toplam kolesterol, HDL-K ve VLDL-K'nin azaldığı, AST ve ALT'nin yükseldiği gözlenmiştir. Sonuç olarak çalışmada; whey proteini, omega-3 yağ asidi ve bariyatrik cerrahinin, yüksek fruktozlu mısır şurubundan kaynaklanan obezite ve dislipidemi üzerindeki olumlu etkisi gösterilmiştir. Elde edilen bu sonuç ile, whey proteini, omega-3 yağ asidi desteği ve bariyatrik cerrahinin birbirine üstünlüklerinin olmadığı saptanmıştır.The aim of this study is to examine the effects of whey protein and omega-3 fatty acid supplementation and Roux-en-Y gastric bypass procedure on body weight loss, histopathological changes in internal organs and biochemical changes in rats made obese by giving high fructose corn syrup. Within the scope of the study, 28 Wistar albino female rats were used and randomly divided into four groups. All rats were made obese by adding 20% high fructose corn syrup-55 to their drinking water for eight weeks. All rats were considered morbidly obese because there was more than 40% weight gain. After the development of obesity, drinking water was given to the first group (control group), whey protein (0.5 g/kg/day) to the second group, omega-3 fatty acid (300 mg/kg/day) to the third group, and after Roux-en-Y gastric bypass drinking water to the fourth group by oral gavage method for 4 weeks. At the beginning of the study, after the development of obesity and at the end of the study, glucose, total cholesterol, HDL-C, VLDL-C, AST, ALT, and uric acid analyzes were performed by taking 1 mL of blood from each rat. At the end of the study, liver, kidney, and pancreas of each rat were taken and evaluated histopathologically. There was no significant difference in body weight between groups. After the intervention, it was observed that total cholesterol, HDL-C, VLDL-C decreased, and AST and ALT increased in the other groups except the control group. As a result, in the study; the positive effects of whey protein, omega-3 fatty acids and bariatric surgery on obesity and dyslipidemia caused by high fructose corn syrup have been demonstrated. With this result, it was determined that whey protein, omega-3 fatty acid support and bariatric surgery were not superior to each other.
