Identification and in vitro bioactivity screening of some marine-derived fungi from Turkish coastlines

Abstract

The marine environment represents a great variety of conditions of pressure, temperature, salinity, and light depending on depth, latitude and interactions with the continental shelf. These extreme and variable conditions enforce marine-derived microbial communities to produce a variety of structurally complex secondary metabolites which may have considerable pharmaceutical potential. In this study four marine-derived fungi were successfully isolated and identified from marine invertebrates (Axinella cannabina, Aplysina aerophoba, Axinella verrucosa, Dendrectilla tremitersis) and investigated for their antioxidant, cytotoxic, antifungal and antimicrobial activities. Three marine fungi strains - Aspergillus sp., Trichoderma sp. and Rhizopus oryzae - were isolated and identified genotypically. Aspergillus sp. had the highest antioxidant activity, and together with Rhizopus oryzae also showed significant cytotoxic activity against HCT-116 cell line. Moreover, among the tested marine-derived fungi extracts Aspergillus sp. was exhibited the highest antifungal activity against Botrytis cinerea.Résumé : Identification et criblage in vitro du potentiel thérapeutique de certains champignongs d’origine marine des côtes turques. L’environnement marin représente une grande variété de conditions de pression, température, salinité et lumière selon la profondeur, la latitude et les interactions avec le plateau continental. Ces conditions extrêmes et variables ont conduit les communautés microbiennes marines à produire une profusion de métabolites secondaires structuralement complexes pouvant avoir un potential pharmaceutique considérable. Dans cette étude, quatre champignons ont été isolés et identifiés à partir d’invertébrés marins (Axinella cannabina, Aplysina aerophoba, Axinella verrucosa, Dendrectilla tremitersis) pour étudier leurs propriétés antioxydantes, cytotoxiques, antifongiques et antimicrobiennes. En particulier, des lignées associées à Aspergillus sp., Trichoderma sp. et Rhizopus oryzae ont été isolées et caractérisées génétiquement. Alors qu’Aspergillus sp. possède la plus forte activité anti-oxydante, ce champignon et Rhizopus oryzae ont également un effet cytotoxique significatif sur la lignée cellulaire humaine HCT-116. De plus, parmi les différentes souches de champignon testées, Aspergillus sp. présente également les activités anti-fongiques les plus élevées contre Botrytis cinerea.
