Erkek infertilitesinde semende adam2(fertilin) catsper ve SOX5 ekspresyonları ve sperm fonksiyonları ile ilişkisi

Abstract

İnfertilite(kısırlık) toplumun %15'ini etkileyen bir durumdur. Çiftlerin yaklaşık %15'i infertilite tanısı ile tedavi olmaktadır.Bu çiftlerde erkek infertilitesi oranı %50'leri bulmaktadır.Erkek infertilitesinde idiopatik sebepler haricinde üzerinde durulan konu sperm yüzey proteinleridir.Spermatozoon membranı oositi çevreleyen membrana yapışma özelliğinin yanısıra, spermin genetik materyali ile membran yapısı arasında belirli bir ilişkinin bulunması açısından da önemlidir. Eğer genetik materyaldeki hatalar spermatozoon morfolojisindeki bozulma ile açıklanabilirse, bilinmeyen erkek infertilite sorunlarının da açıklanabilir duruma gelebileceği düşünülmektedir. Spermatozoon membran ve flagellum (kuyruk) üzerinde bulunan proteinler spermin oosit membranına tutunmasına ve ekstrasellüler matriks alanına girmesine yardımcı olmaktadırlar.Fertilin,disintegrin protein ailesi içinde yer alan,spermatozoon yüzeyinde bulunan, heterodimerik bir proteindir. Tetraspain protein ailesi üyesi olan CD9 ile birlikte, spermatozoon-oosit plazma membranlarının füzyonuna katılmaktadır. Catsper, heterotetramerik 4 ayrı por formlu ? altünitesinden oluşmuş Ca+2 kanalıdır. Sperm flagellum (kamçı) plazma zarında bulunur ve Ca iyonlarının sperm flagellar sitoplazmaya girişini sağlamada görevlidir ve hücre içi pH artışı ile tetiklenmektedir. Sox5 ise post-mayotik germ hücreleri ile round spermatidlerde yüksek seviyede bulunan, gen ekspresyonunu düzenleyen bir transkripsiyon faktörüdür. Catsper promoter bölgesi üzerinde Sox genlerinin bağlanma bölgesi bulunmaktadır ve Catsper gen ekspresyonunu kontrol eden bir konumdadır.Bu çalışmada 30 Normozoospermi ve 30 Oligoastenoteratozoospermi tanısı konmuş 60 kişi ile çalışılmıştır. Sperm motilitesi ve fonksiyonu üzerinde etkili olan fertilin, catsper ve Sox5 protein ekspresyonlarını değerlendirilmiştir. Bu bilgiler ışığında daha sonra yapacağımız çalışmalarla, tespit edilen bu proteinlere karşı antikorlar oluşturularak bu proteine bağlanmaları ve dolayısı ile proteinin aktivitesini inhibe etmeleri sonucunda erkekte doğum konrolünde (kontrasepsiyon) kullanılan bir yöntem olarak katkı sağlamayı amaçlamaktayız.Infertility is a condition that affects 15% of the population. Approximately 15% of the couples are treated with the infertility diagnosis. The rate of male infertility is 50% in couples who cannot have children. Apart from idiopathic causes, sperm surface proteins are another reason for male infertility.The proteins on spermatozoon membrane and flagellum (tail) help to attach to sperm to oocyte membrane and to enter extracellular matrix area. Specifically, spermatozoon membrane is important in terms of having a specific relationship between genetic material of spermatozoon and membrane structure, as well as adhesion properties to the membrane which surrounds oocyte. If defects in genetic material can be understood by deterioration of spermatozoon morphology, it is thought that unknown male infertility problems can be explained by studying spermatozoon membrane.Therefore, this study was conducted on 30 patients with normozoospermia and 30 patients with OAT. We evaluated expressions of Fertilin, Catsper and Sox5 proteins, which are effective on sperm motility and function.Fertilin from the disintegrin protein family is a heterodimeric protein located on surface of spermatozoon. Tetraspain is involved in fusion of spermatozoon-oocyte plasma membranes with CD9 a protein family member. Catsper is a heterotetrameric Ca+2channel composed of 4 subtypes of pore-shaped ? subunits. The sperm flagellum is located in plasma membrane and is responsible for the entry of Ca ions into sperm flagellar cytoplasm and is triggered by intracellular pH increase. Sox5 is a transcription factor that regulates gene expression at high levels in post-meiotic germ cells and round spermatids. There is a binding region of the Sox genes on the Catsper promoter region and Catsper is in a position to control gene expression. Together with current and future researches, we intend to establish a method for controlling fertility in a man by binding present proteins to antibodies, and inhibiting present protein activities.
